Gating strategy for the determination of intracellular PF4 in MDC and PDC. Representative FACS diagrams are presented. Mononucleated cells were identified by their forward/sideward properties (A) and subsequently analyzed with regard to the expression of lineage markers such as CD3, CD14, and CD19 (B). The lineage-negative cells were further tested for expression of CD11c (C) and CD303 (D). Then the PF4 content of the CD11c-positive (E) and CD303-positive cells (F) was evaluated using histogram analysis. The surface expression of HLA-DR was measured accordingly.
