MicroRNA-34a induction during cisplatin treatment of BUMPT cells. (A) BUMPT cells were treated with 40 µmol/L cisplatin for indicated time periods. Total RNA enriched in microRNAs was then isolated for Northern blot analysis using a radiolabeled probe against mature miR-34a. 5S RNA was shown as a sample loading control. Left panel, Northern blots; right panel, densitometry analysis. For densitometry, the miR-34a signal of each condition was normalized with its 5S-RNA signal and then compared with that of the control (0 cisplatin), which was arbitrarily set as one. (B) BUMPT cells were untreated or treated with 40 µmol/L cisplatin for 12 h. Total RNA enriched in microRNAs was then isolated for real-time PCR analysis by using specific primers for mature miR-34a. As an endogenous control, snoRNA-202 was also analyzed in all samples for normalization. Data: mean ± SD, n = 3; *, significantly different from the untreated group.
