Iloprost and treprostinil enhanced IL-10- and poly I:C-induced IL-10 expression and suppressed poly I:C-induced TNF-α expression in human mDCs. Isolated mDCs were pretreated with iloprost or treprostinil for 24 or 48 h, or for 2 h, and then stimulated with poly I:C for 6, 24 or 48 h. Supernatants were collected for cytokine measurement. Iloprost alone (10−8 to 10−7 mol/L) (A) or in the presence of poly I:C (10−9 to 10−7 mol/L) (B) enhanced IL-10 expression in human mDCs. Treprostinil alone (10−9 to 10−7 mol/L) (C) also enhanced IL-10 expression. Iloprost (10−8 to 10−7 mol/L) (D) and treprostinil at higher concentration (10−6 mol/L) (E) suppressed poly I:C-induced TNF-α expression. Results presented are the mean ± SD of six independent experiments using mDCs from six subjects. *P < 0.05 compared with vehicle-treated cells (A, C) or poly I:C-treated cells (B, D, E).
