GNMT deficiency suppressed MOG-specific T-cell responses in peripheral lymphoid tissue. (A) The recovered lymphocytes from draining LNs of 9-d-postimmunized mice were restimulated with various concentrations of MOG35–55 peptide for 24 h. The proliferative response was determined by (3H)thymidine incorporation as described in Materials and Methods. (B and C) Lymphocytes were recovered from draining LNs of 9-d-postimmunized mice and stimulated with PMA and ionomycin in the presence of monensin. The percentages of Th1 and Th17-producing CD4+ T cells were analyzed by flow cytometry. Representative flow cytometric analysis (B) and the average percentages of CD4+ IFN-γ+ and CD4+ IL17A+ T cells (C). All data are presented as mean ± SEM from three mice in each group.
