Simvastatin inhibits TLR8 signaling in HEK Blue TLR8 cells, which is not reversed by treatment with GGPP. (A) HEK Blue TLR8 cells (which stably express TLR8 and a reporter gene expressing a SEAP under the control of a NF-κB/AP-1-inducible promoter) were cultured for 30 min in the presence of 10 µg/mL simvastatin ± 10 µmol/L GGPP, before stimulation with 5 µg/mL R-848 for 16 h. Supernatants were analyzed for SEAP activity by using the Quanti-Blue assay and cell viability (B) was measured by MTT assay. Data are representative of three independent experiments.
