PTX3 deletion does not per se influence macrophage ability to recognize and phagocytose apoptotic cells. (A) The ability of macrophages propagated from bone marrow precursors of WT or PTX3−/− mice was assessed by flow cytometry using two different phagocytes: apoptotic cell ratios are expressed as percentage of phagocytized cells. Results (mean values ± s.e.m) are representative of three independent experiments performed in triplicate. (B) Representative dot plots of WT and PTX3−/− CD11c+ macrophages (y axis) challenged at 37°C (upper panels) and at 4°C (lower panels) with CMTMR (x axis). Expression of (C) MerTK, (D) Gas6, (E) CD163 and (F) MFG-E8 assessed by RT-PCR in WT and PTX3−/− quadriceps at d 1, 3, 7, 10 and 15 after injury.
