TGFβ-induced actin polymerization and induction in αSMA expression are ROCK-dependent. (A) Lens explants were treated with TGFβ (4 ng/mL) in the presence and absence of Y-27632 (20 µM) for 72 h. Fixed explants were costained for F-actin and αSMA and mounted in medium with DAPI to colocalize nuclei. (B) Western blots of pooled lens explant lysates (n ≥ 5 explants per sample) were probed for αSMA, with GAPDH as loading control. (C) Densitometric quantification of αSMA/GAPDH protein indicating the fold reduction in αSMA expression normalized to TGFβ-treated explants.
