Localization and internalization of WT and mutant hETB receptors in CHO cells. hETB was detected using a specific antibody (30). The cells were fixed in 4% paraformaldehyde and permeabilized with 50% ethanol. (A,B) WT, (C,D) G57S, (E,F) R319W, and (G) P383L. For internalization, the cells were treated with 102 6 M ET-1 for 30 min at 48C before fixing and treatment. (B) WT, (D) G57S, and (F) R319W. Control reactions performed using untransfected CHO cells showed no staining (data not shown).
